Three settings determine whether the Amex payment button appears on the checkout page:

Your plan must include the “Pay with Amex” option
Check this by contacting sales administration.

Your store must be linked to an Amex contract
Check your store’s settings from your Expert Back Office (Settings > Shop menu > MID association tab.
If no Amex contract appears in the available contracts or associated contracts, contact American Express to sign up for their service.
Once you have your Amex contract number, contact sales administration to add it to your store.

Certain data sent during payment prevents the Amex button from appearing
Check with your developer to ensure that the card type is not forced during payment.
